Abstract:

Background: The Connective Tissue Growth Factor (CTGF) gene encoding an extracellular matrix (ECM)-associated protein and as a member of the CCN family of proteins plays a major role in fibrosis, inflammation and connective tissue remodeling in a variety of diseases including cancer. The CCN proteins are multifunctional and are involved in cell proliferation, adhesion and cell development during pathogenesis. The purpose of our study was to determine the expression level of CTGF mRNA in patients with colorectal cancer (CRC). Methods: paraffin embedded tissues from 122 patients with colorectal cancer and their matched normal colon tissues and 20 paraffin embedded  normal tissues blocks from normal individual were collected from Taleghani and Shohada hospital in Tehran. The fold change of CTGF expression was analyzed by quantitative real time PCR method, using REST and Prism softwares. Also, LinReg PCR software was also used for the efficiency of primers. Results: We found that expression of CTGF gene in tumor tissue was significantly higher than in adjacent normal tissue (P<0.0001, RQ>2). A Significant increase was also observed in the expression of this gene in right-sided colon cancer than left side. Conclusion: The findings indicated that the expression pattern of CTGF gene is significantly altered in   the development of cancer from normal tissue. Thus, according to major role of CTGF gene in fibrosis, inflammation and connective tissue remodeling, it may be effective in various diseases process including cancer.

Investigating FGF11 gene transcription level in cancer cells among colorectal cancer patients

Background: Colorectal cancer (CRC) is the fourth leading cause of cancer-caused death around the world. Reports of the unnecessary transcription of family genes of the fibroblast growth factor in several types of cancer indicate the role of these factors in tumorgenesis and progression of cancer. Therefore, the level of FGF11 transcription was evaluated in colorectal cancer tumor tissues relat...

CTGF and MLH1 Gene Expression Levels in Colorectal Cancer Tumor Tissues and Adjacent Normal Tissues in Patients in Golestan Province

Background and purpose: Colorectal cancer is the third most common type of cancer in terms of incidence and the second most common cause of cancer-related death worldwide. The aim of this study was to investigate the expression of CTGF and MLH1 gene in colorectal cancer tumor tissues and adjacent normal tissues in patients in Golestan province. Materials and methods: In this experimental study...

Visfatin level in patients with colorectal adenoma

Background: Visfatin is an adipocytokine secreted by visceral adipose tissue. It has been shown that adipocytokines may contribute to the induction of carcinogens and progression of tumors. Previously, we found a significant increase in the visfatin serum level in colorectal cancer patients. Herein, we investigated if this cytokine increases in patients with colorectal adenoma as a precursor of...

Expression of Aldehyde Dehydrogenase 1 in Patients with Colorectal Cancer in North of Iran

Background and purpose: High cancer related mortality can be due to the presence of cancer stem cells (CSCs). Studies suggest aldehyde dehydrogenase (ALDH1) as a marker of CSCs. The aim of this study was to investigate its expression in colorectal cancer and also its association with clinicopathologic characteristics. Materials and methods: In a retrospective study, patients with colorectal ca...

Expression of CTGF and Cyr61 in colorectal cancer.

BACKGROUND The CCN genes encode secreted extracellular matrix proteins cysteine rich-61 (Cyr61), connective tissue growth factor (CTGF) and nephroblastoma overexpressed (Nov). They are involved in diverse cellular functions. Expression of these factors in tumours has produced conflicting results. More recently, research has focused on molecular biomarkers to indicate progression of a disease or...

Investigating the methylation status of DACT2 gene and its association with MTHFR C677T polymorphism in patients with colorectal cancer

Colorectal cancer (CRC) is one of the common causes of cancer death in Iranian population. Both genetic and epigenetic changes have been implicated in CRC pathogenesis. DACT2 gene as one of the WNT signaling pathway inhibitor was shown to display tumor suppressor activity in many cancers. The aim of present study was to investigate the methylation status of DACT2 gene and its ...
